Key fob or inside electric button.... all doors lock or unlock electronically except the driver's door. Driver's door will lock electronically but not unlock. I have to put the key in and turn it manually.

Anyone know what the fix is and how much it should cost to repair?

Edit: 2016 Limited, not that I think that matters.

2"CB shackles, bushings and 1 shim, lockerdown console vault, rear folding seat back mod, snugtop shell with opening side windows, 46g fuel tank, SDHQ sliders and hidden winch, Decked Drawers, Alcan leaf springs and got rid of the interior chrome
I would start with rebooting the truck by disconnecting the battery for 10 minutes.
Then, try a fresh battery in the key fob. These are the first things to start with.

Probably a failing actuator. Cheap and easy to replace. Some replace the motor - time consuming and tedious, albeit cheaper. I can do a door lock actuator in about 10-15 minutes after all four of my 4runner actuators failed with a few months of each other.

Sounds like an actuator. I've had to replace 2 of mine so far and will soon need to do the third. There are a couple options, buy a new one from the dealer and pay $2-400. There are several rebuilt or after market ones that sell for a lot cheaper $30-50 range. I bought some electric motors off of Amazon and replace them for $5 a piece. It's kind of a pain, but when my first one went out, I couldn't find any other options that were under about $200 to fix it.
The actuators are a well known failure point.

Okay guys thank you. I'm not really capable of taking the door apart nor do I have a garage to do it in so I guess I'll probably bring it to the dealer. But, the main thing now is I know it's an actuator and should cost a few hundred bucks to replace. That's extremely helpful.

If you take it to the dealer, I've heard $4-500 with labor. It really is a very simple fix if you buy a replacement. Screw driver, socket (I forget what size) and a torx head driver is all you need to fix it. There are like 2 screws holding the door panel on , one bolt to loosen the window track and then 3 torx bolts holding the actuator on. If you can turn a screw driver, it should really only take you about 20 minutes to do. There are videos on youtube on how to do it. They only thing you have to remember is to make sure the lever for the locking mechanism goes back in the right place. Very simple, just have to remember to do it.

First one for me was covered u fee warranty at the dealership; bill showed $465. The second went out after warranty - did it myself for the $42 aftermarket part. Easy enough. Probably a 2/10 difficulty

Okay well I think the dealer would be a rip off so I ordered an OEM actuator on eBay for $60 and I'll have a local mechanic install it. Like I said, I just don't have a garage to do it in.

    Weird.

The part I ordered on eBay came yesterday. Was going to call a local shop to install it today. Last night, the door lock started working again just fine.

Yeah, I made an appointment to bring it in and then it suddenly started working on its own again before my appointment. Cancelled the appointment and the problem never came back!!!

My driver side actuator went out. It worked, then it stopped working, then it worked until it stopped working. Anyways, don’t order from car parts .com. 1st one I got was broke. Sent it back. 2nd one was broke. Sent it back. 3rd one was the wrong one. Sent it back. Got one from the dealership. $200 and something. It was the right one and not broke.
